As you seen on virustotal, there is no virus. You can send it there too. I don't know why your computer behaves this way. It should be resolved with the lates antivirus definition updates.

I have no idea. VirusTotal does not show anything, Microsoft does not show anything and my Defender is also ok ...
You have to add an exception in the settings of Defender

I install freeware and then try to update to latest version still microsoft defender showing alert that i mention above.

I scanned my uploader and nothing there.
Maybe the file is being infected while downloading to your computer.
Can you upload the file and send it to me? I will analyze it. Maybe you should scan your whole computer.

There is no virus. Just false positive (incorrect) warning. I reported it to those companies
